NeoC101™: A Trojan Molecule for 24-Hour Glow

For decades, cosmetic formulators have struggled to balance the limitations of two well-known actives: vitamin C and azelaic acid. Native vitamin C is highly unstable and difficult to deliver into skin due to its hydrophilicity, while lipophilic derivatives often remain trapped in the stratum corneum. At the same time, azelaic acid suffers from poor solubility and irritation concerns effective concentrations.

NeoC101™ (Diascorbyl Azelate), developed by CORUM, introduces a new co-drug delivery approach designed to overcome these challenges. By linking two ascorbic acid molecules with one azelaic acid, this amphiphilic structure functions as a “Trojan Molecule,” enabling efficient penetration, improved stability, and controlled release within the skin.

With an optimized Log P of -0.14, NeoC101™ achieves a balanced polarity that allows it to pass through the skin barrier without being trapped, while maintaining sufficient affinity for deeper epidermal layers. Once delivered, the molecule undergoes in situ cleavage, releasing vitamin C and azelaic acid in a controlled manner, supporting sustained biological activity where it matters most.

This mechanism is supported by both ex vivo and in vivo data. Studies using the EpiTRI® 3D epidermis model demonstrate the formation of a skin reservoir, with no early-stage permeation detected and a gradual release over 24 hours. In vivo confocal Raman spectroscopy further confirms targeted delivery to approximately 120 μm, corresponding to the dermal-epidermal junction, along with synchronized transformation into active molecules.

Through this efficient delivery system, NeoC101™ demonstrates enhanced performance across multiple pathways. It shows nearly two times stronger antioxidant activity compared to native vitamin C, significantly improved tyrosinase inhibition, and substantially enhanced activity against C. acnes relative to azelaic acid. In addition, it supports collagen-related gene expression and helps reduce UV-induced inflammatory responses, contributing to overall skin resilience and radiance.

From a formulation perspective, NeoC101™ is highly compatible with polyol systems and is particularly suited for waterless formulations, offering improved stability while minimizing common issues such as oxidation and crystallization. With a 100% Natural Origin Index (ISO 16128) and compliance with COSMOS, Vegan, NaTrue, and Halal standards, it aligns with the growing demand for high-performance, sustainable cosmetic ingredients.
